Constitutive generator of Beta-Galactosidase + J23100

This part consitutively expresses lacZ (BBa_I732005) under the control of Ptet strong promoter (BBa_R0040), which is always active because tetR gene (BBa_C0040) does not have any promoter upstream and so it cannot repress BBa_R0040.

If this part is contained in BBa_J61002 vector, RFP (BBa_E1010) is constitutively expressed by BBa_J23100 strong promoter.

This part confers the abilities of metabolizing lactose to lacZ mutant strains (e.g. TOP10 - BBa_V1009) and of glowing (when the vector is BBa_J61002), in order to (indirectly) detect bacterial growth by measuring RFP when the medium is turbid and the optical density (OD) cannot be effectively used as a cell count measure.
