NarP Gene

Lambert iGEM's NarP system is modeled after E.coli’s natural nitrite and nitrate sensor: the Nar Operon, which includes the NarP response regulator, the gene listed above, and NarQ, the membrane-bound sensor protein. The NarP/NarQ system detects both nitrite and nitrate. This specific part is the isolated NarP gene.

The listed NarP’s role within Lambert iGEM’s NarP System, <a href="https://parts.igem.org/Part:BBa_K3411020">BBa_K3411020</a>, is controlled by the promoter <a href="https://parts.igem.org/Part:BBa_J23106">BBa_J23106</a>, a constitutive Anderson Promoter, which initiates the production of TetR. TetR represses the PtetO promoter, producing NarP. In the presence of both nitrate and nitrite, NarQ phosphorylates NarP. Therefore, phosphorylated NarP measures both nitrite and nitrate levels. By using the corrected difference, our NarP and NarL (Lambert iGEM’s improved part of HKUST-Rice 2015 <a href=”https://parts.igem.org/Part:BBa_K1682018”>BBa_K3411020</a>) composite parts are able to evaluate the nitrite levels.

Lambert iGEM plans on cloning and characterizing our NarP part in the coming year as a part of our two-year project, AgroSense.
