NarP Gene
Lambert_GA iGEM 2020's NarP system is modeled after E.coli’s natural nitrite and nitrate sensor: the Nar Operon, which includes the NarP response regulator, the gene listed above, and NarQ, the membrane-bound sensor protein. The NarP/NarQ system detects both nitrite and nitrate. This specific part is the isolated NarP gene.
The listed NarP’s role within Lambert_GA iGEM 2020's NarP System,BBa_K3411020, is controlled by the promoter BBa_J23106, a constitutive Anderson Promoter, which initiates the production of TetR. TetR represses the PtetO promoter, producing NarP. In the presence of both nitrate and nitrite, NarQ phosphorylates NarP. Therefore, phosphorylated NarP measures both nitrite and nitrate levels. By using the corrected difference, the NarP and NarL (Lambert_GA iGEM 2020's improved part of HKUST-Rice iGEM 2015 Part:BBa_K1682018 composite parts are able to evaluate the nitrite levels.
Lambert_GA iGEM 2020 plans on cloning and characterizing the NarP part in the coming year as a part of the two-year project, AgroSense.
