PputA
PputA is the inducible promoter.We screened the whole-genome transcriptome data by setting the conditions that E5_FPKM>E0_FPKM*10 and E0_FPKM < 150, and selected the significant differentially expression genes which express during the early stage of bacteriophage invasion, such as dadA and lldP. We selected the promoter of PputA as our phage-induced promoter in the early stage because the PputA has the maximun value of log2(E5_FPKM/E0_FPKM),which means the PputA has the most significant differentially expression during the early period.
Usage and Biology
PputA is the inducible promoter that When E.coli is infected by a phage, both report and respond gene circuits ((BBa_K3137013)(BBa_K3137014)) are induced to express at the same time. When a phage infects Escherichia coli into the incubation period (about 5 min), PputA will induce the bacteria to express resistant proteins((BBa_K3137006)(BBa_K3137005)) and display green fluorescence.
