Barnase composite part with amber codons
This part contains the RNase Ba (Barnase) (EC 3.1.27) under control of the inducible araBAD promotor (BBa_K808000). It codes for an 12 kDa microbial ribonuclease which cleaves single stranded RNA. Barnase is found in Gram-positive bacteria Bacillus amyloliquefaciens.
Usage and Biology
The coding sequence contains two amber codons at amino acid postitions Q4Am and D46Am. The sequence is inverted (reverse). This composite part can be used to design a possible negative selection plasmid. Sequence and Features
