P2 promoter with RBS
This part is the promoter, P2 with BBa_B0034, a useful RBS. The agr P2 operon is an autocatalytic sensory transduction system in Staphylococcus aureus. P2 promoter regulates the synthesis of AgrB (a transmembrane protein) and AgrD (precursor of AIP autoinducing peptide). AIP binds to AgrC, a membrane binding receptor and it phosphorylates AgrA. The phosphorylated AgrA increases the activity of agr P2 promoter about 50-fold. The cell density at which the agr system is activated is determined by the AgrA-independent activity of P2 promoter The basal agr P2 promoter activity is high enough to make stochastic effects negligible. Design
At first, we wanted to construct two systems which can detect pathogenic bacteria, the one is for Gram-negative bacterium, and the other one is for Gram-positive bacterium. The promoter P2 is related to quorum sensing system of most Gram-positive bacterium, but the sequence on 2016 DNA Distribution Kit Plates is "Inconsistent", so we decided to do a synthesis. We designed the sequence using SnapGene with BBa_B0034, a useful RBS, and added BioBrick Prefix and Suffix, then sent the designed sequence to Sangon Biotech®, a company who has cooperated with our lab for many years to have this part synthesized.
The sequence synthesized was cloned into plasmid pUC-57. The length of this part with BioBrick prefix and suffix is 151bp, so there's no matter with the synthesized part, and the sequencing is all right.
