AatA from Acetobacter acetii

An ABC transporter for the acetogenic bacteria Acetobacter acetii. This transporter binds ATP and moves acetate across the membrane out of the cell. The protein has been shown to confer resistance to acetic acid.

Qdai 2022's Characterization

This transporter contributes to raising the pH of the culture fluid. In our experiments, this part was assembled to E.Coli, BL21 strains with poxB (BBa_K4206001) , which catalyzes the decarboxylation of pyruvate to make acetate ion and CO2. The genetically modified E.coli are cultured and induced with IPTG. After 12 or more hours, we measured the pH of the culture fluid based on the pH test paper. The measurement result was pH 8-9, both with and without IPTG induction. Therefore, this transporter transports acetate ions, whether the ion indicating alkalinity is abundant or not in the cell. Considering the growth limits of E.coli, this transporter needs attention.
