LacZ (RBS+Freiburg standard)

This lacZ gene is derived from the Bacillus reporter vector pAC6. It is constructed in the Freiburg standard with RBS.

lacZ is a β-galactosidase which cleaves the substrates X-Gal (5-bromo-4-chloro-indolyl-β-D-galactopyranoside) and ONPG (ortho-Nitrophenyl-β-galactoside).

lacZ under the control of Pspac in pSB1C3. Plate was induced with xylose.

This BioBrick was used for promoter evaluation in B. subtilis.

We used this lacZ gene to evaluate our promoters PliaG, PliaI, Pveg in pSBBs4S with β-galactosidase-assays for B. subtilis. This gene also works in E. coli which was qualitively seen by blue colonies on LB-Agar-plates that contain X-Gal.
