This biobrick is MESA (Modular Extracellular Sensor Architecture) Part created by combining BBa_K4803005(CD4 signal), BBa_K4803002(gp130 extracellular domain), BBa_K4803006(CD28 Transmembrane domain), BBa_K2969000(TEV protease).
Usage and Biology
This Part is based on the mechanism of MESA, a synthetic receptor. When extracellular IL-6 is detected by this Part and BBa_K4803102, the two Parts dimerize. Upon dimerization, TEV Protease of this Part is activated, which cleaves the intracellular TEV Protease cut site of BBa_K4803102 and releases the lower tTA. The protein is then transported to the endoplasmic reticulum by BBa_K4803005, a CD4-derived ER trafficking signal, and is transmembrane by BBa_K4803006, a CD28-derived transmembrane domain.
