Bacillus subtilis L-arabinose induced killswitch
This composite construct features two distinct systems, the first is meant to allow for the expression of the araR protein, a repressor from Bacillus subtilis 168 that only allows for transcription in the presence of L-arabinose. The first system is composed of the native promoter – paraR (BBa_K4633004), followed by the native ribosomal binding site (RBS) (BBa_K4633005) and the protein araR (BBa_K4633003). Downstream this system is the T1 terminator (BBa_B0010) to ensure that the two systems won’t interfere with one another.
The second system is the inducible system, it is composed of paraE (BBa_K4633006), a promoter containing araR binding sites and thus can be inhibited by araR, its native RBS (BBa_K4633007) and lastly MazF (BBa_K302033), the chosen toxin, meant to induce bacterial demise.
