AND gate - Hypoxia promoter and Lactate operator
This part consists of a hypoxia-inducible promoter (BBa_K387003) intercalated between lactate operators (BBa_K1847008). The hypoxia promoter works under the hypoxic environment, and it has a much higher efficiency of starting transcription than under normal 20% oxygen pressure. The operator regulates the expression of llPRD operon and is involved in L-lactate metabolism. The promoter is repressed by a dimer of LldR, possibly by forming a DNA loop that does not allow the RNA polymerase to bind to the promoter. LldR can also have a function as an activator [1]. The repression of the promoter is removed by lactate, as it binds to LldR and prevents the looping of DNA.
