pPsiR from Sinorhizobium meliloti
This part is the promoter region (0.4 kb upstream) of the PsiR gene of Sinorhizobium meliloti (SMb20483)
pPsiR is predicted to be repressed by the PsiR transcription factor (BBa_K2448007) which is inhibited in the presence of D-Psicose [1].
Usage and Biology
Our team used this pPsiR promoter together with the Sinorhizobium meliloti PsiR transcription factor (BBa_K2448008) to build a Psicose biosensor (BBa_K2448031). The results show that pPsiR is functional in E. coli and that it behaved as predicted under and without induction.
Using the BPROM webserver [2], the predicted features of this promoter are: Threshold for promoters - 0.20 Number of predicted promoters - 1 Promoter Pos: 317 LDF- 3.58 -10 box at pos. 302 gactatcat Score 53 -35 box at pos. 281 ttgcac Score 33 Oligonucleotides from known TF binding sites: lrp: TTTCTTTT at position 273 Score - 9 ompR: TTCTTTTT at position 274 Score - 8
