pPsiA from Sinorhizobium fredii
This part is the promoter region (0.4 kb upstream) of the PsiA gene of Sinorhizobium fredii (NGR_b11530)
pPsiA is predicted to be repressed by the PsiR transcription factor (BBa_K2448007) which is inhibited in the presence of D-Psicose [1].
Usage and Biology
Our team used this pPsiA promoter together with the Sinorhizobium fredii PsiR transcription factor (BBa_K2448007) to build a Psicose biosensor (BBa_K2448028). The results show that pPsiA is functional in E. coli and that it behaved as predicted under and without induction.
Using the BPROM webserver [2], the predicted features of this promoter are: Threshold for promoters - 0.20 Number of predicted promoters - 1 Promoter Pos: 235 LDF- 1.88 -10 box at pos. 219 cgatagtct Score 37 -35 box at pos. 197 ttgacg Score 60 Oligonucleotides from known TF binding sites: argR2: GCAAATAA at position 244 Score - 6
