Banana odor biosynthetic system

We assembled a complete banana odor biosynthetic system (BBa_J45900) by combining the isoamyl alcohol (BBa_J45400) and banana odor generators (BBa_J45200). BBa_J45900 catalyzes the conversion of the cellular metabolite leucine to isoamyl acetate or banana odor.

Usage and Biology

To construct a complete banana odor biosynthetic system, we needed to design an additional biosynthetic device for banana odor precursor production. We designed a second biosynthetic device for production of isoamyl alcohol from the cellular metabolite leucine (BBa_J45400) based on the BAT2 and THI3 genes from Saccharomyces cerevisiae. BAT2 encodes a cytosolic branched-chain amino acid transaminase that catalyzes the conversion of leucine to α-ketoisocaproate. THI3 encodes an α-ketoisocaproate decarboxylase that catalyzes the conversion of α-ketoisocaproate to 3-methylbutanal. 3-methylbutanal is reduced to isoamyl alcohol by an NADH-dependent reaction by an alcohol dehydrogenase. We anticipated that the final reduction step would be carried out by the cellular chassis. Indeed, cultures of the banana odor generator (BBa_J45200) supplemented with 3-methylbutanal produced banana odor.
