Dismed2EnvZ chimeric sensor of two component system

Dismed2EnvZ is a chimeric sensor part of a two-component system (TCS). In specific, The TCS consists of a chimeric sensor formed by Dismed2 that is, the extracellular domain of RetS, which is a sensor protein from P. aeruginosa known to sense mucin (1). The transmitter domain of the chimeric sensor is composed of HisKA and HATPase domains EnvZ. EnvZ is part of the popular E. coli-derived two-component system EnvZ-OmpR (2,3). The EnvZ-OmpR TCS functions in a such way that EnvZ, when activated, phosphorylates the transcription factor OmpR, which subsequently activates the OmpC promoter. In our design OmpC regulates the transcription of the essential gene gmk, making our organism dependent on mucin.
