RNA Linker binding MS2 Coat Protein and Antitermination Protein N
This part will self-cleave when transcribed resulting in an RNA consisting of an MS2 hairpin (BBa_K3113006) and a BoxB hairpin(BBa_K3657015).
Usage and Biology
MS2 hairpin is specifically bound by the MS2 Coat Protein (BBa_K3218006). The BoxB hairpin is specifically bound by Antitermination protein N (BBa_K3657014). Consequently this part can be used to link two proteins of interest fused respectivly to the MS2 Coat Protein and to the Antitermination protein N.
It was planned to test the ability of this linker to bring proteins together by using MS2 Coat Protein/Antitermination protein N fused to Split-GFP subunits. The following parts were planned to be used: BBa_K3657030, BBa_K3657031.
Due to the COVID19 pandemics no experimental data about this part was obtained

Hammerhead ribozymes are sequences that cleave themselves when transcribed. They were introduced order to ensure that the transcript does not include artifacts from promoter or terminator. Those sequences could interfere with the secondary structure of the RNA.
