NarL Gene

Lambert_GA iGEM 2020's NarL system is modeled after E.coli’s natural nitrate sensor, the Nar Operon, which includes the NarL response regulator, and NarX, the membrane-bound sensor protein. The NarL/NarX system detects nitrate. This specific part is the isolated NarL gene.

Description

The listed NarL’s role within Lambert_GA iGEM 2020’s (improved from HKUST-Rice 2015, BBa_K1682018) system serves as an acceptor of the phosphate group from NarX. The promoter BBa_J23106, a constitutive Anderson Promoter, initiates the production of TetR. TetR then represses the PtetO promoter. By regulating the amount of aTc, the molecule that inhibits TetR, the team can then control levels of NarL. In the presence of nitrate, NarX phosphorylates NarL. Then, phosphorylated NarL binds to a specified promoter and correlates to measured nitrate levels. Further details about the continued expression of the NarL part are listed in the description HKUST-Rice provides on their part. Therefore, phosphorylated NarL measures nitrate levels.

Lambert_GA iGEM 2020 will continue cloning and begin characterization of our NarL part in the coming year as a part of our two-year project, AgroSense.
