NarP Gene
Lambert iGEM's NarP system is modeled after E.coli’s natural nitrite and nitrate sensor: the Nar Operon, which includes the NarP response regulator, the gene listed above, and NarQ, the membrane-bound sensor protein. The NarP/NarQ system detects both nitrite and nitrate. Similar to the NarL system, the promoter BBa_J23106, a constitutive Anderson Promoter, initiates the production of TetR, which represses the PtetO promoter, producing NarP. NarP is phosphorylated by NarQ in the presence of both nitrate and nitrite. Therefore, phosphorylated NarP indirectly measures both nitrite and nitrate levels. By using the corrected difference, our NarP and NarL composite parts are able to evaluate the nitrite levels. This specific part is the isolated NarP gene.
Lambert iGEM plans on cloning and characterizing our NarP part in the coming year as a part of our two year project, AgroSense.
