Part:BBa_K2810000
WRKY intron for plant-targeted RNAi constructs
This part contains the sequence of the WRKYintron, which is commonly used as to separate the sense and anti-sense parts of an RNAi contruct for gene silencing. This has been used many times by plant biologists and is part of the pJawohl8-RNAi binary plasmid.
The WRKYintron is cloned into the the golden gate version of pSB1C3 (P10500) using BsmBI and conforms to the [http://2016.igem.org/Resources/Plant_Synthetic_Biology/PhytoBricks Phytobrick standard]. It is flanked by BsaI sites together 5' CCAT and 3' AATG cloning sequences for subsequent cloning into a level 1 plasmid.
This has been used to generate the composite RNAi constructs detailed in the 2018 Cardiff_Wales Parts page
