Promoter for Bacillus megaterium+RBS
This part is a combination of promoter and RBS. We got the sequence of RBS (BBa_B0030) from its part page, and combined it to the promoter got from the plasmid PHIS1525. This plasmid can be used in Bacillus magaterium for the expression of foreign proteins. Then the whole part has been synthesized by AuGCT. Bacillus magaterium is a kind of potential genetic engineering host, which is now frequently used in industry and academic fields. It has lots of advantages over another host as following: 1. It is able to yield exceptionally high-level foreign protein. 2. Its heredity is stable 3.The foreign proteins can be secreted out of the cell, in that case, we just need to collect the supernatant rather than lyse the cell by sonication. In a word, it can be easily operated. 4 Endotoxin free for Bacillus magaterium, which is eligible for biosafety requirement. However, there are few iGEM teams using these strain as their host, so we are going to be one of the pioneers and we try to find several types of the promoter, RBS and terminator and plan to make combinations between them. Our final purpose is to find the most effective way for protein expression in Bacillus magaterium for further uses.
