NarL Gene
Lambert iGEM's NarL system is modeled after E.coli’s natural nitrite and nitrate sensor: the Nar Operon, which includes the NarL response regulator, the gene listed above, and NarX, the membrane-bound sensor protein. The NarL/NarX system detects both nitrite and nitrate. This specific part is the isolated NarL gene.
The listed NarL’s role within Lambert iGEM’s (improved from HKUST-Rice 2015, BBa_K1682018) NarL System. The promoter BBa_J23106, a constitutive Anderson Promoter, which initiates the production of TetR. TetR represses the PtetO promoter, producing NarP. In the presence of both nitrate and nitrite, NarQ phosphorylates NarP. Therefore, phosphorylated NarP measures both nitrite and nitrate levels. By using the corrected difference, our NarP and NarL (Lambert iGEM’s improved part of HKUST-Rice 2015 BBa_K1682018 composite parts are able to evaluate the nitrite levels.
Lambert iGEM plans on cloning and characterizing our NarL part in the coming year as a part of our two-year project, AgroSense.
