scaffold of sgRNA in CRISPR/Cas system

A simplified version of the type II CRISPR/Cas9 system from Streptococcus pyogenes, which only consists of the Cas9 nuclease and a chimeric single guide RNA (sgRNA), is currently the most popular genome editing tool. The sgRNA, which harbors a 20 nt protospacer that specifically recognizes and binds to the target sequence, and a 42 nt structural Cas9-binding handle, directs Cas9 to the host genome, and creates a DNA double-strand break (DSB). Deriving from CRISPR/Cas9, the nuclease-deficient Cas9 (dCas9) coexpressed with a sgRNA, has been established for efficient gene repression. This part is the 42 nt structural dCas9-binding handle.
