siRNA from 5' region of Myzus persicae SP3 secreted protein
This part is taken from the 5' region of the SP3 gene, from the aphid Myzus persicae , and expressed in Nicotiana benthamiana to test its effects on the aphid. The hope is that it will prevent formation of aphid bacteriocytes and thus prevent their symbiotic relationship with Buchnera aphidicola , killing the bacteria and the host.
SP3 is a predicted secreted protein that localizes to [http://rspb.royalsocietypublishing.org/content/280/1750/20121952 aphid bacteriocyte cells].
This sequence is taken from the coding strand from a 5’ region of the Myzus persisae SP3 gene. The Cardiff_Wales team is attempting to target this gene using RNAi.
SP3_5_5 is cloned into the golden gate version of pSB1C3 (P10500) using BsmBI and conforms to the [http://2016.igem.org/Resources/Plant_Synthetic_Biology/PhytoBricks Phytobrick standard].
It is flanked by BsaI sites together 5' TACT and 3' CCAT cloning sequences for subsequent cloning into a level 1 plasmid.
This sequence is incorporated into a level 1 plasmid for plant expression as part of an RNAi construct with the complimentary sequence found in BBa_K2810008
