NarP Gene
NarP is a part of the Nar Operon which detects both nitrate and nitrite. Our system is modeled after E.coli’s natural nitrite sensor. Similar to NarL system, the promoter BBa_J23106, a constitutive Anderson Promoter, produces TetR, which represses the PtetO promoter, producing NarP. The NarP gene is phosphorylated in the presence of both nitrate and nitrite. Therefore, when NarP eventually phosphorylates PdcuS, which is further explained in the NarP composite Part:BBa_K3411020, it is measuring the levels of both nitrate and nitrite. By using the corrected difference our NarP and NarL composite parts, we are able to evaluate the levels of Nitrite. This specific part is the isolated NarP gene.
Lambert iGEM plans on further characterizing our part in the coming year as a part of our two year project, AgroSense.
