PglcF

PglcF is an inducible promoter.We screened the whole-genome transcriptome data by setting the conditions that E20_FPKM>E5_FPKM*10, 0.8<E0_FPKM/E5_FPKM<1.2 and E5_FPKM<150, and selected the significant differentially expression genes which express during the later stage of bacteriophage invasion, such as gabT and gabP. We selected the promoter of PglcF as our phage-induced promoter because the PglcF has the maximun value of log2(E5_FPKM/E0_FPKM)and the minimun value of log2(E5_FPKM/E0_FPKM), which means the PglcF has the most significant differentially expression during the later period and the least significant differentially expression during the early stage.

Usage and Biology

PglcF is the inducible promoter that works when phages infect bacterial for around 20 min. If the resistant protein (BBa_K3137006)(BBa_K3137005) successfully resists phage infection, the inducible promoter PglcF will not induce the expression of downstream gene. If the resistant protein cannot kill phages, the phages will continue to infect. When phages infect bacteria into the burst period (about 20 min), the inducible promoter PglcF will induce expression of the red fluorescent protein gene mCherry(BBa_K3137004) and the downstream toxic protein gene (BBa_K628000), which will make the bacteria display red fluorescence and lyse cells before the assembly of phages.
