This system designed by the MIT 2006 iGEM team generates mint smell by biosynthesizing salicylic acid from chassis-produced metabolites (chorismate) and then converts salicylic acid into methyl salicylate (wintergreen scent).  It uses a stationary phase promoter followed by an inverter (see <bbpart>BBa_J45996</bbpart> for details) to only express the necessary enzymes during exponential phase.

To construct a complete wintergreen odor biosynthetic system, we needed to design an additional biosynthetic device for wintergreen odor precursor production.  We designed a biosynthetic device for production of salicylate from the cellular metabolite chorismate (<partinfo>BBa_J45320</partinfo>) based on the ''pchBA'' genes from ''Pseudomonas aeruginosa''.  The enzyme PchA, an isochorismate synthase, catalyzes the conversion of chorismate to isochorismate.  The enzyme PchB, an isochorismate pyruvate-lyase, catalyzes the conversion of isochorismate to salicylate and pyruvate.  Serino ''et al.'' previously demonstrated salicylate formation in ''E. coli'' using the ''pchBA'' genes.

Exponential phase wintergreen odor biosynthetic system

We assembled a complete, exponential phase dependent wintergreen odor biosynthetic system (BBa_J45800) by combining the salicylic acid (BBa_J45320) and exponential phase dependent wintergreen odor generators (BBa_J45181). BBa_J45800 catalyzes the conversion of the cellular metabolite chorismate to methyl salicylate or wintergreen odor. Wintergreen odor should only be produced in exponentially growing cells.

Usage and Biology

To construct a complete wintergreen odor biosynthetic system, we needed to design an additional biosynthetic device for wintergreen odor precursor production. We designed a biosynthetic device for production of salicylate from the cellular metabolite chorismate (BBa_J45320) based on the pchBA genes from Pseudomonas aeruginosa. The enzyme PchA, an isochorismate synthase, catalyzes the conversion of chorismate to isochorismate. The enzyme PchB, an isochorismate pyruvate-lyase, catalyzes the conversion of isochorismate to salicylate and pyruvate. Serino et al. previously demonstrated salicylate formation in E. coli using the pchBA genes.
