crp_trigger_4_5’trigger_phi29 (BBa_K4438907) is a single-stranded DNA having 11 nucleotides. Figure 1A) shows the secondary structure and minimum free energy.

Usage and Biology

The part crp_trigger_4_5’trigger_phi29 (BBa_K4438907) has complete complementarity to the part Aptamer_crp (BBa_K4438900) and blocks the 5’ region of the CRP aptamer. Figure 1D) Shows the secondary structure of both parts hybridised at 37° Celsius. CRP binds with Aptamer_crp (BBa_K4438900) and displaces the crp_trigger_4_5’trigger_phi29 (BBa_K4438907) [1]. This part has complete complementarity with part crp_Target_4(BBa_K4438908). Phi 29 DNA extension polymerase extends the template strand and in-vitro transcription of the duplex forms multiple broccoli light-up aptamers. Different levels of CRP can be detected using all these three parts.
