Split-GFP N-Terminal Subunit

This part is the sequence for the N-Terminal subunit of the Split-GFP. It is used with the C-Terminal subunit of the Split-GFP (BBa_K3657027).


Usage and Biology


Split-GFP was designed based on the Green Fluorescent Protein (GFP). It is made of two subunits. Both subunits do not emmit fluorescence by themselves. When the subunits are brought in close proximity of each other, they bind and form a fluorescent complex. If both subunits are co-expressed in one cell without an agent that brings them together, no fluorescence is observed.


Split-GFP is mostly used to prove interaction between two proteins. To achieve that one should fuse proteins of interest with respectivly N- and C-Terminal subunits of the Split-GFP. We,iGEM Heidelberg 2020, planned on using the Split-GFP to test the concept of an RNA linker.
