This composite part includes the lasB promoter region BBa_R0079 hooked up to GFP gene from 2004 BBa_E0040. Upstream of that cassette is a constitutive tet promoter that drives production of LasR from Pseudomonas aeruginosa BBa_C0179. In an E. coli chassis the device was exposed to commercially-available PAI-1 (a small hydrophobic signalling molecule), grown for several hours, then immunoblotted for GFP production (Dundee Figure 1). The device worked as expected and produced detectable levels of GFP over time (Dundee Figure 1).
