This is a short complementary DNA sequence that binds to our 85-mer Cortisol aptamer (BBa_K4693002). Together with the aptamer, it forms an aptasensor that can quantify the neurotransmitter cortisol. This version of the cDNA was designed based on the kD value of the aptamer-cDNA pair compared to the kD value of the cortisol binding with the 85-mer aptamer. Multiple complementary sequences were designed to find a sequence with a kD value higher than -16.1 that would not affect the binding and formation of the FRET system designed. The chosen cDNA had a kD value of -10.3 and even though there were multiple possible structures the conformations did not affect the binding position of the cDNA hence the interaction of the fluorophore quencher pair.
