generate EL222, a kind of photosensitive factor

Usage

When el222 gene is expressed, the photosensitive transcription factor EL222 is generated.The photosensitive factor EL222 comprises two domains: the Light-Oxygen-Voltage(LOV)Sensing domain and the N-terminal domain. Upon stimulation by blue light (450nm), EL222 dimerizes, activating its role as a transcription factor. It promotes the transcription of the Pc120, thereby initiating the expression of the downstream gal4 gene.

Biology

EL222 is a photosensitive transcription factor comprising two functional domains. One is the Light-Oxygen-Voltage (LOV) sensing domain, which responds to light at 450nm, leading to homologous dimerization of EL222. The second domain is the Helix-Turn-Helix (HTH) DNA-binding domain, which specifically recognizes and binds to the Pc120(BBa_K4616005) repeat sequence.

The previously described photosensitive transcription factor (TF) consists of three components: the Nuclear Localization Signal (NLS), the VP16 Trans-Activation Domain, and the bacterial-derived protein EL222[1]. The presence of the LOV domain in EL222 allows the photosensitive transcription factor to activate the transcription of downstream genes regulated by the Pc120 when exposed to blue light (450nm).
