L.reuteri autophagy pathway
This part is used to sense the change of chloride ion concentration which will cause diarrhea, then turn on the autolytic gene and convey the signal to the downstream E. coli engineering bacteria. When the chloride concentration goes up, N-acetylmuramidase gene acmA will activated by promoter Pgad, then lysis of the cell wall of gram-positive bacteria. Besides, gene gadR and rrnB is added into this system to regulates the expression of AcmA.
In this parts, rrnB is an RNAseH II that specifically degrades the ribonucleotide moiety on RNA-DNA hybrids in Lactococcus lactis. pgadR, gadR and Pgad is a series of gene that regulates the Cl−-dependent expression of AcmA positively.
