Yeast Promoter - Medium Strength Transcription.

Usage and Biology

This part is the yeast promoter pHTB2, which is a medium strength yeast promoter taken from the MoClo YTK toolkit.

This promoter part is part of a larger library of BioBrick_BASIC_YeastPromoters we have designed as part of iGEM 2020 offering a range of different transcription strengths (See BBa_K3559003 through K3559009). This will allow teams to choose promoters based on desired levels of expression or to build large combinatorial libraries.

Characterisation

As a result of the Covid pandemic we have been unable to characterise or use this part as of yet, we therefore make no statement on the performance of this part. Having said that, pHTB2 is well-characterised as a part of the MoClo Yeast Toolkit.

Whilst pHTB2 is well-characterised in the MoClo YTK paper (https://doi.org/10.1021/sb500366v), we have added the BioBrick and BASIC prefixes and suffixes which may have an impact on gene expression levels compared to what is seen in the paper. Additionally, during BASIC cloning relatively large linkers are added between each part, this will additionally change the context of this promoter and may have effects on its strength.

This part is surrounded by both BASIC iP/iS and BioBrick prefix and suffix, allowing users of this part to carry out cloning using either the BASIC or BioBrick methodologies.
