Part:BBa_K3016500
Human growth hormone gene with 6xHis
This part includes human growth hormone encoding gene (hGH) with 6xHis tag in the C-terminus.
Usage
This part can be used to express hGH that can be relatively easily purified because of the hexa His tag.
Biology of the human growth hormone
hGH is a peptide hormone that stimulates growth, cell reproduction, and cell regeneration in humans, and is essential in human development. It also stimulates the production of IGF-1 and increases the concentration of glucose and free fatty acids in plasma.
hGH is a 191-amino acid, single-chain polypeptide. It is synthesized, stored and secreted by somatotropic cells located in the lateral wings of the anterior pituitary gland. hGH has a molecular weight of 22,124 Da. The protein structure includes four helices that interact with the GH receptor. The genes that encode human growth hormone are growth hormone 1 (somatotropin; pituitary growth hormone) and growth hormone 2 (placental growth hormone; growth hormone variant), and they are located in the q22-24 region of chromosome 17.
Clinical significance
A recombinant form of hGH is used to treat growth disorders and growth deficiencies. These include Turner syndrome, Prader–Willi syndrome, and chronic renal failure.
