RnaG120-based NOT gate of MlcRE
RnaG120 is a short noncoding RNA (sRNA) complementary to icsA mRNA 5' UTR that induces stem loop formation in icsA mRNA 5' UTR leading to transcription termination in Shigella flexneri. We employ RnaG120/icsA 5' UTR as efficient NOT-gate for MlcRE to create a glucose-induced regulator.
Gene of interest (GOI) mRNA is preceded by icsA 5' UTR and inverted MlcRE. Constitutive GOI transcription is driven by Anderson promoter BBa_J23106. Without glucose, MlcRE induces transcription of RnaG120, which induces terminator function of icsA 5' UTR upstream of the GOI coding sequence. Upon repression by glucose, MlcRE no longer induces RnaG120 transcription. icsA 5' UTR terminator is no longer formed, and the GOI coding sequence is transcribed.
