Part:BBa_K2572034
TlpA39-mWasabi
This is a composite part of the thermosensitive repressor Tlp39 and the monomeric florescent protein reporter GFP. TlpA39 is from Salmonella typhimurium, quite similar to TlpA36 (BBa_K2447012).It is a temperature sensitive protein which is capable of binding to promoter pTlpA. Under temperature of above 39 degrees Celsius, TlpA39 protein would stay in monomeric form and hence it loses the ability to bind to the promoter pTlpA39, the downstram mWasabi is not repressed, and florescence is emitted; If the temperature drops below 39 degree Celsius, TlpA39 proteins would form dimers; which are capable of repressing the pTlpA promoter and prevent the recruitment of RNA polymerase to repress downstream gene expression, mWasabi will be repressed, and no florescence will appear. It therefore functions as a reporter of temperature. In our project, 39 degrees Celcius is the threshold for our recombinant E. coli to produce anti-inflammatory drugs.
The part was characterized by us as effective and no leakege at 36 degrees Celsius was detected.
