Composite brick encoding LovTAP hybrid protein under Lacl promoter

Improved LovTAP encoding brick allowing compatibility with all assembly standards. Based on the original LovTAP encoding biobrick BBa_K191006 designed by Le Thanh Tu Nguyen from École Polytechnique Fédérale de Lausanne 2009 iGEM team.

Part BBa_K2330002 improves on the original brick's versatility of use by optimizing the coding sequence for the iGEM registry while retaining the original translated sequence. By retaining original translated sequence the optimized brick BBa_K2330002 should not experience any decrease in activity.


Usage and Biology

LovTAP is a fusion protein consisting of a light sensitive Light-oxygen-voltage-sensing (LOV) domain and tryptophan repressor domain (TAP). The LOV domain shares a high sequence identity nonphototropic hypocotyl 1 (NPH1-1) a blue light receptor found in Avena sativa while the trp-repressor (trpR) is found in E. coli. The fusion protein operates as a DNA binding protein mediated by blue light (470 nm). Photoexcitation at 470 nm and subsequent activation of the LOV domain results in the conformational change of the trpR domain allowing DNA binding, DNA disassociation occurs when the LOV domain returns to the inactive dark state causing the conformation to revert.

To determine if the adaptions made to the LovTAP biosensor had an impact to its efficiency testing was carried out similarly to the original LovTAP biobrick. Lovtap under the lacl promoter was induced using IPTG and the fusion proteins activity was determined using reporter system BBa_K191005 also designed by Le Thanh Tu Nguyen from École Polytechnique Fédérale de Lausanne 2009 iGEM team.
