RFP and Tetracycline resistence(tetA) under the control of a modified lacZ promoter; Fusion protein

TetA and RFP under the control of an optimized PlacZ combined with a generator of the fusion protein SH2:cMyc:cI(434)- BBa_K2082237

The main function is the expression of the fusion protein SH2:cI(434), which contains the DNA binding domain (cI) fused with the SH2 domain as target of the designed positive control of the two-hybrid system. Together with BBa_2082221 the proteins HA4:RpoZ and SH2:cI(434) are interacting through affinity of HA4 and SH2 (Wojcik et al. 2010). After all the high affinity of these proteins can be used to increase the activation rate of the reporter gene coded on BBa_K2082237. As reporter genes the tetracycline resistence tetA and the fluorescence gene of RFP are implemented. Therefore it is possible to make screening and selection bacterial two-hybrid experiments with this created part.
