GoldenGate compatible AmilGFP, Yellow Chromoprotein
Variation of the part BBa_K592010 that has been made compatible with the GoldenGate MoClo Assembly Standard as it is free from BsaI and BpiI recognition sequences.
iGEM14_NRP-UEA-Norwich chose to use this part as one of their reporters as the chromoprotein naturally exhibits strong yellow color when expressed. The construct was cloned to be part of a transcriptional unit with MAS as the promoter and octopine synthase (Agrobacterium tumefaciens) as the terminator.
The next stage of The Green Canary Project would be to use this part alongside the part AmilCP (BBa_K1467201) as the two reporters in a multi-gene system with the aim of producing a plant that turns yellow on activation of the promoter BS3, induced by the pepper pathogen (TALE) AvrBS3. (Similarly, the blue chomoprotein would be expressed on activation of the promoter Xa27 by the TALE AvrXa27). This would allow identification of the presence of this pathogen in the area.
