GoldenGate compatible AmilCP, Blue chromoprotein
A variation of the part BBa_K592009 that has been made compatible with the GoldenGate MoClo Assembly Standard as it is free from BsaI and BpiI recognition sequences.
iGEM14_NRP-UEA-Norwich chose to use this part as one of their reporters as the chromoprotein naturally exhibits strong blue colour when expressed. The construct was cloned to be part of a transcriptional unit with monnopine synthase (MAS) as the promoter and octopine synthase (ocs) as the terminator.
The next stage of The Green Canary Project would be to use this part alongside the part AmilGFP (BBa_K1467202) as the two reporters in a multi-gene system with the aim of producing a plant that turns Blue on the activation of the promoter Xa27, induced by the rice pathogen (TALE) AvrXa27. (Similarly, the Yellow chomoprotein would be expressed on activation of the BS3 promoter by the TALE AvrBS3).
