T7.2 gene 1
Encodes gp1: T7 RNA polymerase from wild-type T7; Suppressor mutation in 19 allows no gene 1 late in infection. It's thought that gene 1 is hanging out at the CJ terminator at the pause site... recruiting 18 and 19 after packaging. (Studier FW, PC)
Usage and Biology
Approximately at position 2500 hundred is a possible E. coli promoter (S1). TTGAC is the consensus -35 region located at 2498-2502 the possible -10 region is TGTACT from 2520-2525
The GTTGAC from 2497-2502 encodes V-D dipeptide. Redesigned to be GTAGAT (~same tRNA abundance)
The GAT peptide from 2518-2520 an D amino acid is mutated to GAC to disrupt the -10 region.
Nucleotides 4-39 were redesigned to encode the same amino-acid sequence as the original sequence, but lead to minimal secondary structure with the preceding RBS (BBa_Z0262) in the T7.2 design. RBS-CDS junction is predicted to have ΔG = -0.01 kCal/mol.
