We met some problem in our construction. It seems that the overexpression of repressor protein has cytotoxicity which made troubles in our process. When we ligated tetR protein coding sequence in our circuit as the picture followed .Then we fund that recombination happened without Cre recombinase. So the promoter had been removed. It seems that the overexpression of repressor protein has cytotoxicity which made troubles in our process. Compared with the random mutation, the lox71 site is easier to delete the promoter and the bacteria without tetR protein can get a growth advantage. So, at last, there is only the bacteria without promoter in the medium.
