Time-Delay Input Filter
The first component in this part is an inducible promoter. This acts as the receiver for the input signal and can be modified according to the need of a system. In the sequence submitted, this is the Lac Operon Promoter. The promoter regulates the transcription of a weak inducer protein that must homodimerize in order to become active. The need for homodimerization introduces the time delay as the protein needs to build up for an extended period of time before becoming active. The exact protein chosen for this component can be varied depending on the delay needed. In the sequence submitted, this is HD-Zip family transcription factor Hat4. This transcription factor is followed by a transcriptional terminator sequence and then the promoter which our transcription factor activates; in this case COX5B-1. This secondary promoter directly regulates the part's output. This output is modular but in the submitted sequence it is GFP.
