Type II Secretion of Chromate and Uranium Reductase with TorA
This device is composed of a promoter, TorA signal peptide tag which a Silver-fusion compatible BioBrick part that can be attached to other proteins to target those proteins for export out of the cytoplasm and targets folded proteins to the Tat-dependent pathway of Type II secretion, Silver-fusion compatible chromate and uranium reductase coding sequence and two transcriptional terminators. The reason why we try to secrete the ChrR30 protein is that the reductase activity for both uranium and chromate is improved when the cell extracts were used instead of whole cell. Barak et al. (2006) found out that the dissepearance of chromate and uranium caused by cell extracts is much higher than the one caused by whole cell. Thus, we decided to secrete our reductase enzyme to increase the efficiency of the bioremediation process. Please consult the original pages for each of these parts for more information.
