FseI restriction endonuclease actuator construct
This composite part includes a strong bicistronic RBS, the FseI protein coding gene, and a pT-T7 terminator.
Usage and Biology
The coding region produces the restriction enzyme FseI. As FseI is produced, it cleaves DNA at recognition sites using exonuclease activity. This can be utilized for experiments by surrounding a gene of interest with FseI cut sites. Under the control of a promoter of choice, FseI can be expressed and cut out either the promoter or entire gene in the gene of interest. This way, one can control temporally the knock out of a gene of interest.
