Part:BBa_M10037

Designed by: Patrick Harrigan   Group: Berkeley BioE140L - S09   (2009-05-10)

{˂KILR˃}

This basic part codes for an alpha helix which forms coiled-coil heterodimers with the alpha helix protein EILD. This part can be used in conjuction with EILD for binding based purification methods or or for binding based localization of proteins. This part includes no start and no stop codon and is meant to be used in translational fusion peptides.

The sequence was designed using a multi-state computational method, which attempted to optimize the free energy of KILR/EILD heterodimer formation relative to KILR and EILD homodimer formation. The calculated free energy change from the homodimeric state to a KILR/EILD heterodimer is -8.7 kcals/mol. Using the well characterized homodimeric coiled coil GCN4 as a template, the sequence design attempted to create coiled-coil forming proteins which bind preferentially to form heterodimers. This part is in BglBricks Standard. It is flanked by BamHI and BglII sites instead of XbaI and SpeI. More information about the BglBricks Standard is available at:
