Part:BBa_M10036
{a~TraA˃}
This part codes for a C-terminal display protein derived from the TraA gene (sequence at [http://www.ncbi.nlm.nih.gov/sites/entrez?Db=gene&Cmd=retrieve&dopt=full_report&list_uids=1263548&log$=databasead&logdbfrom=nuccore NP_061453]) located on the F plasmid. The preprocessing sequence has been removed using the tools found at [http://phobius.cbr.su.se/ Phobius]. The f pillin subunit coded for by the sequence allows for the formation of an f pillus in F+ strains of E. coli. F pilli are recognition sites for infectious bacteriophages such as fl, fd and M13. Proteins fused to the C terminus of traA display on f pilli and block the recognition mechanism necessary for infection by these bacteriophages. In bacteria displaying antigenic peptides on f pilli, the ability to infect is recovered in bacteriophages displaying a specific recombinant antibody fragment. This can be used in protein engineering systems as only bacteriophages displaying the ligand for the protein displayed on traA will infect the bacteria amplifying the signal of ligand binding ([http://www.ncbi.nlm.nih.gov/sites/entrez PMID: 9356245]).
This part is in BglBricks Standard. It is flanked by BamHI and BglII sites instead of XbaI and SpeI. More information about the BglBricks Standard is available at:
