Strain E-15 EG3

Function: The strain is capable of utilizing ethylene glycol as a sole carbon source, and grows optimally at 30 mM of ethylene glycol media.
Note: The original stab culture was re-streaked on ethylene glycol agar for three generations to ensure degradation activity.

Media Components (per liter):

  • 34 mM NaH2PO4
  • 64 mM K2HPO4
  • 20 mM (NH4)2SO4
  • 1 μM FeSO4
  • 0.1 mM MgSO4
  • 10 μM CaCl2
  • 30 mM Ethylene Glycol

    For Agar plates (per liter):
    Add in the above components and add 15g of bactoagar to achieve a 1.5% (w/v) mixture.

    Growth Rates: (at 37°C, at 150rpm)
  • In 30 mM ethylene glycol media: around 24 hours to reach OD 0.2
  • In 30 mM ethylene glycol agar: around 84 hours to develop single colonies.

    Experimental Results:

    When Strain E-15 EG3 was repassaged in new media, we saw a general increase in growth rate over time. We performed two trials simultaneously on the same original strain, labeled Tube 1 and Tube 2. For Tube 1, we saw a 74.8% increase and for Tube 2, we saw a 227.84 % increase, both over a time period of 285.95 hours.
