ZCD with Histidine Patch Thioredoxins and Hist-tag
This is the functional unit which makes ZCD soluble in E. coli. It includes a inducible promoter, mutated thioredoxin to increase solubility, the ZCD ORF, a hist-tag for easy purification and finally a double terminator. This part has been shown to highly express ZCD in E. coli. It is suggested that this part be used in a plasmid also containing araC for the optimal repression of this protein before the time of induction.
The information that lead to the development of the this vector tag with ZCD came out of the following paper:
Bouvier, Florence et al. “Oxidative Remodeling of Chromoplast Carotenoids : Identification of the Carotenoid Dioxygenase CsCCD and CsZCD Genes Involved in Crocus Secondary Metabolite Biogenesis.” 15.January (2003): 47–62.
