GFPT1

This part includes two repeats of a 20 bp sequence. This sequence is complementary to a section of the genomic GFP coding sequence in E.coli Keio strain BW31005 (CGSC#8185). Topoisomerase binding sites -CCCTT- have been placed at the beginning and end of each 20 bp sequence allowing Topoisomerase to nick after the last thymine, bind to the 3' phosphate, and peal off a 20 bp ssDNA probe. This part should be paired with GFPT2, a biobrick that works in the same fashion as GFPT1, but recognizes and hybridizes with the 20 bp GFP genomic sequence 10 bp downstream from the GFPT1 hybridization site.
